The Barkindji Biosphere Reserve provides outstanding opportunities to undertake long-term manipulative and survey research on natural habitats, and the interface between these habitats and urban/agricultural land use. Professor Ary Hoffmann (Centre for Environmental Stress and Adaptation Research, La Trobe University) has developed the following outline for the Barkindji Biosphere research program.

Major problems facing the area are the loss of biodiversity due to grazing and clearing for agricultural purposes, and stress arising from other forms of human impact, including the increasing problem of salinity. Also of significance are the ways in which these problems may affect the general sustainability of agricultural, tourism and urban activities in the area. Particular problems may affect various vegetation types to different extents, but they are prevalent across the whole area, from the flood plains of the Murray to the mallee and chenopod shrublands of the interior.
